A first generation genetic linkage map of the European flat oyster Ostrea edulis (L.) based on AFLP and microsatellite markers ArchiMer

This study presents the first genetic linkage map for the European flat oyster Ostrea edulis. AFLP and 20 microsatellite markers were genotyped in a three-generation pedigree comprised of 2 grand-parents, 2 parents and 92 progeny. Chi-square goodness of fit tests revealed high segregation distortion, which was significant for 32.8% of markers. Sixteen microsatellites and 235 AFLPs (170 AFLPs type 1:1 and 65 AFLPs type 3:1) were used to build sex-specific linkage maps using CriMap software. A hybrid zone of the genus Ctenomys: a case study in southern Brazil Genet. Mol. Biol.

We describe variation at microsatellite loci and the chromosomal polymorphisms of a hybrid population, and hybridizing populations of Ctenomys minutus (the minor tuco-tuco) from the coastal plain of Rio Grande do Sul, southern Brazil. Cytogenetic analysis and a survey of six microsatellite loci included 101 specimens of C. minutus from the parental populations (2n/AN = 42/74 and 48a/76) and their contact zone. Cytogenetic analysis recorded 26 different karyotypes exhibited by 50 individuals from the hybrid population. The objective of this study was to characterize the genetic variability of Anglo-Nubian goats using microsatellite markers. The study was conducted using herds from four municipalities of Central-Northern Piauí (Teresina, José de Freitas, Campo Maior, and Angical), where technical information is scarce. Seven markers suggested by FAO were used (ILSTS11, McM527, INRA23, ETH10, OarfCB304, OarfCB48, and MAF209). The samples were genotyped using a 7% polyacrylamide gel. A simple miniprep based on early elimination of highly acidic and proteinaceous mucilages through ethanol washing of the tissue homogenates has been developed for the extraction of genomic DNA from mature leaves and seeds of Corchorus spp. As compared to high cetyltrimethylammonium bromide (CTAB)-NaCl DNA extraction followed by ethanol-based removal of remnant mucilages from the DNA pellet, this simple miniprep consistently and reproducibly recovers high amounts of DNA with good spectral qualities at A260/A280 and A260/A230. The purified DNA is efficiently digested by restriction endonucleases, and is suitable for PCR amplification of nuclear microsatellites with expected allele sizes.

Hybridization between divergent lineages often results in reduced hybrid viability. Here we report findings from a series of independent molecular analyses over several seasons on four life stages of F1 hybrids between the newts Triturus cristatus and T. marmoratus. These two species form a bimodal hybrid zone of broad overlap in France, with F1 hybrids making up about 4% of the adult population. We demonstrate strong asymmetry in the direction of the cross, with one class (cristatus-mothered) making up about 90% of F1 hybrids. By analyzing embryos and hatchlings, we show that this asymmetry is not due to prezygotic effects, as both classes of hybrid embryos are present at similar frequencies, implicating differential selection on the two hybrid classes...

Olive (Olea europaea L.) production in the world has been made by using many cultivars, and the genetic uniformity of commercial cultivars is important for standard olive oil and table olive production. The genetic variation among and within commonly cultivated olive cultivars in Turkey was analyzed using SSR markers. A total of 135 leaf samples were collected from 11 commonly cultivated olive cultivars from 11 provinces in four geographical regions of Turkey. Characterisation of 22 polymorphic microsatellite loci for the Broadbill Swordfish, Xiphias gladius ArchiMer

The Broadbill Swordfish is harvested by fisheries throughout the world. In order to devise more effective management strategies, we need a clear understanding of the population structure of the species. From a library of 29 tetranucleotide repeats, 22 microsatellite markers were characterised for 94 swordfish samples captured from eastern and western Australia. The number of alleles ranged from 2 to 26 and observed heterozygosity from 0.066 to 0.923. We have identified 18 microsatellites that will be valuable in the examination of swordfish population structure.

ABSTRACT Hilly chickens were characterized from January 2015 to September 2016 considering climate factors (light and heat). The experimental birds were divided into three groups (heat stress; light and control). The heat was generated in the chicken’s shelter by a black shaded light (Lantern) for two to three hours more after sunset. A lighting device (Lantern) was kept in the chicken’s shelter for the same period for increasing daylight duration. Characterization of thirteen new polymorphic microsatellite markers from the honeycomb grouper Epinephelus merra ArchiMer

Epinephelus merra (Serranidae) is the most common and widespread species of this genus, mostly found in reef habitats from South Africa to Central Pacific. Thirteen polymorphic microsatellite loci were isolated and characterized from this species. Polymorphism was studied in two localities from the Comoros archipelago-Moh,li and Moroni. Allelic diversity was 7.5, polymorphism ranging from 3 to 13 alleles. Overall heterozygosity was high with an average observed heterozygosity of 0.587. Three of these loci were characterised by heterozygote deficiencies for both populations. Finally, ten of the 13 developed primers could be used in future population genetics study of E.merra that might be useful in a context of marine biodiversity conservation.

Comparative analysis of microsatellite variability in five macaw species (Psittaciformes, Psittacidae): application for conservation Genet. Mol. Biol.

Cross-amplification was tested and variability in microsatellite primers (designed for Neotropical parrots) compared, in five macaw species, viz., three endangered blue macaws (Cyanopsitta spixii [extinct in the wild], Anodorhynchus leari [endangered] and Anodorhynchus hyacinthinus [vulnerable]), and two unthreatened red macaws (Ara chloropterus and Ara macao). Contrasted levels of genetic diversity in a benthic Mediterranean octocoral: Consequences of different demographic histories? ArchiMer

Understanding the factors explaining the observed patterns of genetic diversity is an important question in evolutionary biology. We provide the first data on the genetic structure of a Mediterranean octocoral, the yellow gorgonian Eunicella cavolini, along with insights into the demographic history of this species. We sampled populations in four areas of the Mediterranean Sea: continental France, Algeria, Turkey, and the Balearic and Corsica islands.
